Sus antecedentes:

  • Las tauropatías implican la agregación de la proteína tau hiperfosforilada, lo que conduce al deterioro cognitivo.
  • La tau patológica se disemina a través de regiones cerebrales y tipos neuronales específicos, pero los mecanismos de vulnerabilidad selectiva no están claros.

Objetivo del estudio:

  • Investigar los mecanismos celulares y moleculares subyacentes a la vulnerabilidad neuronal selectiva en tauropatías.
  • Identificar los actores moleculares clave que contribuyen a la susceptibilidad regional a la patología tau.

Principales métodos:

  • Inmunohistoquímica en tejido cerebral humano post-mortem de tauropatías.
  • Purificación de ribosomas de traducción viral (vTRAP) en ratones P301S (PS19) y controles.
  • Secuenciación de ARN, bioinformática (Gene Ontology, MECT) y caracterización electrofisiológica.

Principales resultados:

  • Las neuronas excitatorias muestran una acumulación predominante de tau en comparación con las neuronas inhibitorias en diversas tauropatías.
  • Las neuronas y regiones vulnerables presentan alteración de la transmisión sináptica y la excitabilidad neuronal.
  • Se identificó Mef2c como regulador de la mielinización y la organización sináptica en regiones vulnerables; hallazgos validados en tauropatías humanas.

Conclusiones:

  • Las neuronas excitatorias son el objetivo principal de la patología tau en las tauropatías.
  • La vulnerabilidad selectiva está relacionada con la alteración de la neurotransmisión y la composición sináptica, potencialmente regulada por Mef2c.
  • Este estudio dilucida los impulsores moleculares de la susceptibilidad regional y celular en la neurodegeneración.
