Sus antecedentes:

  • La enfermedad de Alzheimer (EA) afecta a millones de personas, con causas poco claras para la mayoría de los casos esporádicos.
  • Las características patológicas incluyen ovillos de tau y placas amiloides.
  • Una variante de expansión de repetición (RE) del gen CASP8, CASP8-GGGAGA-AD-R1, se asocia con un mayor riesgo de EA.

Objetivo del estudio:

  • Investigar los mecanismos de la enfermedad de la variante CASP8-GGGAGA-AD-R1.
  • Determinar el papel de las proteínas que contienen CASP8 polyGR en la patogénesis de la EA.

Principales métodos:

  • Transfección de células SH-SY5Y con variantes de CASP8 para expresar proteínas polyGR.
  • Inducción de estrés oxidativo mediante peróxido de hidrógeno (H2O2).
  • Análisis de los niveles de proteína de polyGR, tau fosforilada (p-tau) y caspasas mediante inmunofluorescencia.

Principales resultados:

  • Las células positivas para PolyGR mostraron un aumento de la caspasa 8 (1,9 veces) y p-tau (2,4 veces).
  • El estrés oxidativo aumentó significativamente los niveles de proteína CASP8 polyGR (16 veces) y p-tau (1,5 veces).
  • Los niveles de caspasa-3 no aumentaron de manera similar.

Conclusiones:

  • Los agregados de PolyGR contribuyen a la patología tau y la activación de la caspasa-8 en la EA.
  • El estrés oxidativo promueve la acumulación de polyGR, empeorando las características patológicas de la EA.
