Sus antecedentes:

  • La preservación cognitiva en adultos mayores de alto riesgo sigue siendo poco comprendida.; Las variantes raras (VR) pueden contribuir a la heredabilidad faltante de la enfermedad de Alzheimer (EA).; Las poblaciones fundadoras, como los Amish, pueden enriquecer las VR, facilitando el descubrimiento genético.

Objetivo del estudio:

  • Identificar VR y genes asociados relacionados con la preservación cognitiva.; Aprovechar los datos de secuenciación del genoma completo (WGS) de la población Amish.; Investigar los fundamentos genéticos de la resiliencia cognitiva en el envejecimiento.

Principales métodos:

  • Secuenciación del genoma completo (WGS) de 868 individuos Amish (518 con deterioro cognitivo leve, 350 con deterioro cognitivo).; Pruebas de asociación de VR en todo el genoma basadas en genes que comparan el estado cognitivamente normal (CU) vs. cognitivamente deteriorado (CI).; Análisis de VR codificantes usando SKAT-O y VR no codificantes usando STAARpipeline, teniendo en cuenta las covariables.

Principales resultados:

  • Se encontraron asociaciones sugestivas para VR codificantes en los genes B3GNT9 y RANBP10 (p < 6.59x10-5).; Se identificaron tres VR de sentido erróneo con impacto proteico moderado.; También surgió una señal sugestiva de 14 VR en la región promotora de C1QL4 (p < 7.00x10-6).

Conclusiones:

  • Los análisis de asociación de VR en todo el genoma basados en genes revelaron vínculos sugestivos con la preservación cognitiva en los Amish.; Las VR muestran potencial para desvelar la arquitectura genética completa de la EA.; La investigación adicional sobre estas VR podría identificar nuevas dianas terapéuticas para la EA.
