Sus antecedentes:

  • La enfermedad de Alzheimer (EA) es un trastorno neurodegenerativo progresivo que causa demencia, relacionado con la deficiencia de acetilcolina y el estrés oxidativo.; La investigación de nuevos agentes terapéuticos es crucial para el manejo del declive cognitivo y la neuropatología de la EA.

Objetivo del estudio:

  • Evaluar los efectos anti-Alzheimer de la hordenina en un modelo de rata de demencia inducida por escopolamina.; Evaluar el impacto de la hordenina en marcadores conductuales, neuroquímicos, inflamatorios y de estrés oxidativo en la EA.

Principales métodos:

  • Las ratas fueron tratadas con hordenina (50, 100 mg/kg) o donepezilo (5 mg/kg) durante siete días.; Se administró escopolamina para inducir demencia, seguida de pruebas conductuales y análisis de tejido cerebral para neurotransmisores, acetilcolinesterasa, estrés oxidativo y marcadores inflamatorios.

Principales resultados:

  • La escopolamina indujo déficits cognitivos, aumentó la actividad de la acetilcolinesterasa y elevó los marcadores oxidativos/inflamatorios (LPO, TNF-α, IL-1β, NF-kB).; Los tratamientos con hordenina y donepezilo amelioraron significativamente estos cambios, demostrando efectos neuroprotectores contra la patología similar a la EA.; El tratamiento con hordenina mejoró la memoria y la función cognitiva, redujo la neuroinflamación y atenuó el estrés oxidativo.

Conclusiones:

  • La hordenina exhibe propiedades neuroprotectoras significativas contra la patología similar al Alzheimer inducida por escopolamina en ratas.; La hordenina puede servir como un candidato terapéutico para la enfermedad de Alzheimer al mitigar la neuroinflamación, el estrés oxidativo y el declive cognitivo.; Se justifica una investigación adicional sobre el potencial de la hordenina para revertir los procesos neurodegenerativos en la EA.
