Sus antecedentes:

  • La deposición de tau es una característica distintiva de las enfermedades neurodegenerativas, incluida la enfermedad de Alzheimer (EA).; La patología tau se propaga a través de vías conectadas, con la región de unión a microtúbulos (MTBR) de tau formando semillas.; E2814 es un anticuerpo dirigido contra tau-MTBR en ensayos clínicos para prevenir la propagación de la patología tau.

Objetivo del estudio:

  • Investigar el mecanismo de E2814 y 7G6 en la captación celular de tau.; Evaluar el impacto de estos anticuerpos en la internalización neuronal y microglial de tau.

Principales métodos:

  • Se utilizaron cultivos in vitro de neuronas de roedores y humanas (glutamatérgicas derivadas de iPSC).; Se aislaron y cultivaron microglía primaria y derivada de iPSC.; Se midió la tau intracelular utilizando tau marcada con fluorescencia, anticuerpos e imágenes de células vivas.

Principales resultados:

  • E2814 y 7G6 disminuyeron la captación de tau tanto en neuronas de roedores como humanas.; La captación de tau por la microglía aumentó en presencia del anticuerpo, dependiente de la función efectora.

Conclusiones:

  • E2814 puede impedir la propagación de la patología tau al inhibir la captación neuronal de tau.; El anticuerpo también puede acelerar la eliminación de tau-MTBR a través de vías microgliales.
