Sus antecedentes:

  • La disfunción microglial está implicada en la patogénesis de la enfermedad de Alzheimer (EA).
  • Las terapias existentes dirigidas a la microglía no han producido beneficios clínicos para los pacientes con EA.
  • SIGLEC10, expresado exclusivamente en la microglía cerebral, desempeña un papel en la EA.

Objetivo del estudio:

  • Investigar el potencial terapéutico de ONC-841, un anticuerpo monoclonal anti-SIGLEC10, en modelos de ratón de EA.
  • Evaluar la capacidad de ONC-841 para cruzar la barrera hematoencefálica y dirigirse a SIGLEC10 microglial.
  • Evaluar la eficacia de ONC-841 en la reducción de la patología de la EA y su mecanismo de acción.

Principales métodos:

  • Se desarrollaron modelos de ratón de EA que expresan SIGLEC10 cruzando modelos de EA con ratones transgénicos humanos SIGLEC.
  • Se administró ONC-841 por vía intravenosa y se confirmó la penetración de la barrera hematoencefálica y la ocupación de SIGLEC10 microglial.
  • Se evaluó la acumulación de placas amiloides y pTau, se midieron biomarcadores en plasma y se investigó el mecanismo de acción mediante secuenciación de ARN nuclear única y ensayos de fagocitosis in vitro.

Principales resultados:

  • El tratamiento con ONC-841 condujo a una eliminación significativa de las placas de beta-amiloide en el cerebro.
  • Se observaron niveles reducidos de pTau181 y Tau total en plasma de ratones tratados.
  • ONC-841 mejoró la fagocitosis microglial de agregados proteicos y rejuveneció las funciones microgliales.

Conclusiones:

  • El anticuerpo anti-SIGLEC10 ONC-841 demuestra actividad terapéutica en modelos de ratón de EA.
  • ONC-841 promueve la eliminación de placas amiloides y reduce los biomarcadores de tau al rejuvenecer las funciones microgliales.
  • Estos hallazgos respaldan el desarrollo clínico de ONC-841 como una inmunoterapia dirigida a la microglía para la enfermedad de Alzheimer.
