  • Las terapias para la enfermedad de Alzheimer (EA) tienen un éxito limitado, lo que requiere tratamientos alternativos.; Los cannabinoides, como el cannabidiol (CBD) y la delta-9-tetrahidrocannabinol (THC), poseen propiedades antiinflamatorias y son bien tolerados.; El efecto combinado de CBD y THC sobre la patología tau, una característica clave de la EA, sigue siendo poco comprendido.

Objetivo del estudio:

  • Evaluar los efectos del cotratamiento crónico con CBD y THC en comportamientos relevantes para la enfermedad en ratones hembra TAU58/2 transgénicos.; Investigar el potencial terapéutico de una proporción específica de CBD:THC (50:3 mg/kg/día) en un modelo de ratón de EA.; Explorar el impacto de los cannabinoides en la función motora, la memoria, la ansiedad y la inhibición de estímulos sensoriales en el contexto de la tauopatía.

Principales métodos:

  • Se trataron ratones hembra transgénicos TAU58/2 de seis meses de edad y controles de tipo salvaje con CBD+THC (50:3 mg/kg/día) o vehículo durante cinco semanas.; Se realizaron evaluaciones conductuales, incluyendo función motora, memoria de reconocimiento espacial y social, ansiedad e inhibición de estímulos sensoriales, después de tres semanas de tratamiento.; El análisis de datos se centró en los efectos del tratamiento y el genotipo en diversas pruebas conductuales.

Principales resultados:

  • Los ratones TAU58/2 mostraron déficits en la función motora, la inhibición de estímulos sensoriales, el comportamiento de ansiedad y la memoria espacial.; El cotratamiento crónico con CBD:THC mejoró significativamente la función motora y redujo parcialmente el comportamiento de ansiedad en ratones transgénicos.; Los cannabinoides mostraron una tendencia a mejorar la memoria espacial, aunque no fue estadísticamente significativa; el reconocimiento social y la inhibición de estímulos sensoriales no se vieron afectados.

Conclusiones:

  • El tratamiento a largo plazo con CBD:THC (50:3 mg/kg/día) demostró efectos terapéuticos sutiles pero prometedores en los comportamientos relacionados con la tauopatía en ratones de mediana edad.; Este estudio proporciona la primera evidencia del potencial terapéutico del cotratamiento con CBD:THC para abordar los síntomas conductuales de la tauopatía.; Los hallazgos sugieren que la terapia con múltiples cannabinoides, particularmente proporciones específicas, puede ser prometedora para el tratamiento de la EA, lo que justifica una mayor investigación sobre los mecanismos moleculares subyacentes.
