Sus antecedentes:

  • La enfermedad de Alzheimer y las tauropatías primarias se caracterizan por la agregación de tau.
  • DA-7503 es un inhibidor de molécula pequeña oralmente disponible que se dirige a la oligomerización de tau.
  • Estudios preclínicos en modelos de ratón mostraron una mejora de la memoria y una reducción de la patología de tau.

Objetivo del estudio:

  • Evaluar las propiedades farmacológicas de DA-7503.
  • Evaluar su potencia y especificidad contra isoformas de tau patológicas.
  • Determinar sus efectos terapéuticos sobre los déficits motores y la agregación de tau en el modelo de ratón JNPL3.

Principales métodos:

  • Se probó la potencia inhibitoria contra la agregación de tau 3R y 4R en células HEK293.
  • Ratones transgénicos JNPL3 recibieron administración oral diaria de DA-7503 durante 4.5 meses.
  • Se evaluaron los efectos terapéuticos mediante pruebas de función motora y análisis de agregación de tau.

Principales resultados:

  • DA-7503 demostró una potente inhibición de la agregación de tau 3R y 4R.
  • El DA-7503 oral mejoró significativamente la función motora en ratones JNPL3 en múltiples pruebas de comportamiento.
  • Una dosis de 15 mg/kg redujo los agregados de tau patológicos en el tronco encefálico y la médula espinal.

Conclusiones:

  • DA-7503 restauró eficazmente la función motora y redujo los agregados de tau en un modelo de ratón de tauropatía.
  • El compuesto exhibe una potente actividad contra isoformas de tau patológicas y un perfil de seguridad favorable.
  • DA-7503 es un candidato terapéutico prometedor para la enfermedad de Alzheimer y las tauropatías primarias, actualmente en ensayos clínicos de Fase 1.
