Sus antecedentes:

  • El deterioro cognitivo se deriva de la disfunción de la red, no de problemas sinápticos aislados.
  • La neuroinflamación exacerba el daño neuronal y el deterioro cognitivo.
  • Las terapias actuales se enfrentan a un punto de no retorno a medida que la enfermedad progresa.

Objetivo del estudio:

  • Investigar la reprogramación neuronal de astrocitos tóxicos en neuronas.
  • Disminuir la neuroinflamación y estabilizar la función de la red neuronal.
  • Mejorar finalmente la cognición en un modelo de enfermedad de Alzheimer.

Principales métodos:

  • Expresión de factores de transcripción pronuronales (ASCL-1 o ASCL-1 SA6) en ratas AD TgF344 envejecidas y en camadas de control no transgénicas.
  • Administración de vectores virales (AAV2/5) bajo un promotor GFAP en el hilio del hipocampo.
  • Monitorización conductual y examen patológico 7 semanas después de la inyección.

Principales resultados:

  • Se observó una mejora significativa de la función cognitiva 7 semanas después de la reprogramación neuronal.
  • Reducción de la astrogliosis y la microgliosis, lo que indica una disminución de la neuroinflamación.
  • Aumento de la densidad neuronal y disminución de la carga de placas amiloides.

Conclusiones:

  • La reprogramación neuronal reduce eficazmente la neuroinflamación y mejora la supervivencia neuronal en el hipocampo.
  • El estudio demuestra un retorno a un ambiente cerebral homeostático.
  • Esta estrategia muestra ser prometedora para el tratamiento del deterioro cognitivo en modelos de envejecimiento y AD.
