Sus antecedentes:

  • Alcadeinß (Clstn3) es una proteína de membrana sináptica que se escinde en péptidos, incluidos p3-Alcß37, p3-Alcß40 y p3-Alcß9-19.; Estos péptidos, en particular p3-Alcß9-19, pueden restaurar la función mitocondrial alterada por oligómeros de amiloide-beta en modelos neuronales y modelos de ratón con enfermedad de Alzheimer (EA).; La identificación de los objetivos moleculares de p3-Alcß9-19 es crucial para comprender su potencial terapéutico en la EA.

Objetivo del estudio:

  • Identificar y validar los objetivos moleculares del péptido p3-Alcß9-19.; Analizar los perfiles de expresión génica en los cerebros de un modelo de ratón con EA tratado con p3-Alcß9-19.; Investigar los mecanismos terapéuticos de p3-Alcß9-19 en el contexto de los cambios en la expresión génica relacionados con la EA.

Principales métodos:

  • Administración de p3-Alcß9-19 (1 mg/kg) a ratones AppNL-F/NLF (un modelo de EA) durante un mes.; Análisis de secuenciación de ARN (RNAseq) de regiones cerebrales disecadas de ratones tratados y ratones control (tratados con PBS).; Análisis comparativo de los cambios en la expresión génica en modelos de ratón y pacientes humanos con EA.

Principales resultados:

  • La administración de p3-Alcß9-19 alteró la expresión génica tanto en células neuronales como no neuronales dentro del cerebro del ratón con EA.; Muchos genes que se infraexpresan en pacientes humanos con EA se sobreexpresan o muestran una tendencia a la sobreexpresión en ratones tratados con p3-Alcß9-19.; Por el contrario, algunos genes sobreexpresados en pacientes humanos con EA se normalizaron en el modelo de ratón con EA tras el tratamiento con p3-Alcß9-19.

Conclusiones:

  • p3-Alcß9-19 protege las funciones neuronales modulando los perfiles de expresión génica alterados por la acumulación de proteínas patógenas en el cerebro.; El péptido p3-Alcß9-19 demuestra potencial como agente terapéutico para preservar la homeostasis cerebral y combatir la neurodegeneración.; Los hallazgos preclínicos respaldan la investigación clínica adicional de ALC919, una formulación transdérmica de p3-Alcß9-19, para el tratamiento de la enfermedad de Alzheimer.
