Sus antecedentes:

  • La neuroinflamación impulsada por la microglía es central en la patogénesis de la enfermedad de Alzheimer (EA).
  • Estudios genéticos implican variantes de la fosfolipasa C gamma 2 (PLCγ2) en el riesgo y la protección contra la EA.
  • Las variantes protectoras de PLCγ2 se correlacionan con una mayor actividad proteica, lo que sugiere un potencial terapéutico.

Objetivo del estudio:

  • Identificar moléculas pequeñas que mejoren la expresión o actividad de PLCγ2.
  • Explorar la activación de PLCγ2 como estrategia terapéutica para la EA.
  • Mimetizar los efectos protectores de la variante PLCγ2•P522R.

Principales métodos:

  • Cribado de alto rendimiento (Biblioteca codificada con ADN, Espectrometría de masas de selección de afinidad) para identificar uniones a PLCγ2.
  • Ensayos bioquímicos y celulares (Fluorimetría de barrido diferencial, Ensayo de desplazamiento térmico celular, IP-One, fagocitosis) para la participación del objetivo y la actividad.
  • Estudios de relación estructura-actividad y evaluaciones in vivo para la farmacocinética y el impacto en la neuroinflamación.

Principales resultados:

  • Descubrimiento de nuevos activadores moleculares pequeños de PLCγ2 con optimización preliminar.
  • Demostración de la participación del objetivo, la eficacia bioquímica y la farmacología celular.
  • Análisis in silico sugiere idoneidad para el descubrimiento de fármacos del sistema nervioso central (SNC).

Conclusiones:

  • La activación de PLCγ2 representa una nueva vía terapéutica para la enfermedad de Alzheimer.
  • Se identificaron andamios estructuralmente diversos capaces de activar enzimas y efectos celulares.
  • Se recomiendan moléculas sonda para la validación del objetivo, con el desarrollo de compuestos similares a los líderes para ensayos clínicos.
