Sus antecedentes:

  • El tratamiento de la enfermedad de Alzheimer (EA) tiene necesidades insatisfechas a pesar de las terapias de amiloide.
  • La patología tau se correlaciona fuertemente con el deterioro clínico de la EA.
  • BIIB080, un oligonucleótido antisentido, se dirige al ARNm tau para reducir la producción de proteína tau.

Objetivo del estudio:

  • Evaluar la eficacia, seguridad y tolerabilidad de BIIB080 en la EA temprana.
  • Evaluar la respuesta a la dosis de BIIB080 en la reducción de la patología tau.
  • Investigar el impacto de BIIB080 en el deterioro cognitivo y los biomarcadores de la EA.

Principales métodos:

  • Estudio de Fase 2, aleatorizado, doble ciego, controlado con placebo (CELIA, NCT05399888).
  • 416 participantes con EA temprana (MCI o demencia leve), de 50 a 80 años.
  • BIIB080 intratecal o placebo durante 76 semanas; criterio de valoración principal: cambio en CDR-SB a las 76 semanas.

Principales resultados:

  • Se ha completado la inscripción para el estudio de Fase 2 CELIA.
