Sus antecedentes:

  • Varoglutamstat (PQ912) es un inhibidor oral de la glutaminil ciclasa dirigido a la formación de amiloide-beta (Aβ) y CCL2.
  • Los datos preclínicos y de Fase 1 respaldaron su evaluación en la enfermedad de Alzheimer (EA) temprana.

Objetivo del estudio:

  • Determinar la dosis óptima, segura y bien tolerada de varoglutamstat para la EA temprana.
  • Evaluar la seguridad y eficacia del fármaco durante 72 semanas en un ensayo de Fase 2.

Principales métodos:

  • Los pacientes con EA temprana confirmada por biomarcadores fueron aleatorizados a varoglutamstat o placebo.
  • Un diseño secuencial probó dosis descendentes con análisis de futilidad a las 24 semanas.
  • El punto final primario fue CDR-sum-of-boxes (CDR-SB); los puntos finales secundarios incluyeron medidas cognitivas y funcionales.

Principales resultados:

  • La dosis de 600 mg se seleccionó en base a la seguridad en la Fase 2A; sin embargo, el ensayo se interrumpió prematuramente.
