Sus antecedentes:

  • Sorbs2 es una proteína adaptadora del citoesqueleto en neuronas del hipocampo.; Las variantes genéticas en Sorbs2 están relacionadas con el inicio retrasado de la enfermedad de Alzheimer (EA).; La función precisa de Sorbs2 en las neuronas sigue sin estar clara.

Objetivo del estudio:

  • Investigar el papel mecanicista de Sorbs2 en la función neuronal y la memoria.; Explorar el potencial de Sorbs2 como proteína de ARN (RBP) específica de neuronas.; Evaluar el impacto de la deficiencia de Sorbs2 en el aprendizaje, la memoria y la neurogénesis.

Principales métodos:

  • Se generaron ratones Knockout (KO) de Sorbs2 para análisis conductuales y moleculares.; Se realizaron pruebas de reconocimiento de objetos espaciales (SOR) y condicionamiento al miedo con señales (CFC).; Se realizaron ensayos de estabilidad de microtúbulos, extracciones de proteínas de sinaptosomas y secuenciación de ARN.

Principales resultados:

  • Los ratones KO de Sorbs2 exhibieron un aprendizaje y una memoria deficientes en las pruebas SOR y CFC.; La secuenciación de ARN reveló que Sorbs2 regula vías implicadas en la estabilidad de los microtúbulos y la iniciación de la traducción.; La deficiencia de Sorbs2 provocó una regulación atípica del empalme alternativo, afectando los sitios de inicio de la traducción.

Conclusiones:

  • Sorbs2 desempeña un vital papel en la regulación de la estabilidad de los microtúbulos y el empalme alternativo en las neuronas.; Sorbs2 está implicado en la formación y recuperación de la memoria.; Se desarrollaron herramientas para estudiar la expresión de Sorbs2 y su papel en modelos de ratón de EA.
