Objetivo del estudio:

  • Desarrollar una estrategia novedosa de evolución dirigida para superar las limitaciones en la edición principal mediada por recombinasa de ADN.
  • Diseñar recombinasas de ADN de alta eficiencia que funcionen eficazmente con almohadillas de aterrizaje cortas.
  • Mejorar la eficiencia y especificidad generales de la integración genómica a gran escala.

Principales métodos:

  • Se desarrolló una estrategia de evolución dirigida que utiliza almohadillas de ADN de longitud progresivamente decreciente como presión selectiva.
  • Se cribaron e identificaron variantes de recombinasa evolucionadas con actividad catalítica mejorada.
  • Se validó el rendimiento de las variantes modificadas en conjunción con la edición principal para la integración genómica.

Principales resultados:

  • Las variantes de recombinasa modificadas, VK y AVK, demostraron una actividad intrínseca sustancialmente mejorada.
  • La actividad mejorada se mantuvo en almohadillas de aterrizaje de longitud corta, superando las ineficiencias previas.
  • Se logró un efecto sinérgico con la edición principal, aumentando significativamente la eficiencia de inserción y las tasas generales de integración al tiempo que se preservaba la especificidad genómica.

Conclusiones:

  • Se introdujo una nueva clase de recombinasas de ADN de alta eficiencia (VK y AVK).
  • Se presentó una estrategia de ingeniería robusta con aplicabilidad general para el desarrollo de terapias génicas y la investigación genómica fundamental.
  • Se demostró un avance significativo en la eficiencia y especificidad de la integración genómica a gran escala.
