Sus antecedentes:

  • Los genomas de los mamíferos poseen millones de elementos reguladores que controlan la expresión génica.
  • El consorcio ENCODE había catalogado previamente 0,9 millones de elementos cis-reguladores humanos y 300.000 en ratones.

Objetivo del estudio:

  • Ampliar el registro ENCODE de cCREs utilizando nuevos conjuntos de datos y métodos computacionales.
  • Proporcionar un recurso integral para comprender la regulación génica en diversos tipos de células y tejidos.

Principales métodos:

  • Aprovecho los nuevos conjuntos de datos ENCODE y los enfoques computacionales mejorados.
  • Datos de caracterización funcional integrados de ensayos como STARR-seq, MPRA, CRISPR y modelos de ratón transgénico.
  • Amplió el registro para incluir 2.37 millones de cCREs humanos y 967.000 de ratones.

Principales resultados:

  • El registro ampliado cubre cientos de tipos de células y tejidos, mejorando la comprensión de la regulación genética.
  • Más del 90% de los cCREs humanos fueron perfilados funcionalmente, revelando funciones reguladoras complejas.
  • Se identificaron miles de nuevos cCREs silenciadores, algunos con doble función de potenciador/silenciador.
  • KLF1 fue identificado como un nuevo gen causal para los rasgos de los glóbulos rojos.

Conclusiones:

  • El registro ampliado ENCODE cCRE es un recurso valioso para la investigación en genómica reguladora.
  • La integración con otros datos ENCODE ayuda a interpretar la variación genética y a identificar los genes asociados a los rasgos.
  • Este recurso hace avanzar el estudio del papel del genoma regulador en la salud y la enfermedad.
