Sus antecedentes:

  • Los agentes de contraste clonables como la Proteína Verde Fluorescente (GFP) son vitales para la microscopía de fluorescencia.; Existe una falta de agentes clonables similares para la microscopía electrónica biológica.; Los métodos existentes luchan por proporcionar contraste en todas las escalas de longitud biológica.

Objetivo del estudio:

  • Informar sobre una novedosa NanoPartícula de Selenio clonable (cSeNP) para contraste molecular en imágenes biológicas.; Demostrar la utilidad de cSeNP en múltiples modalidades de imagen, incluyendo microscopía electrónica, microscopía de fluorescencia y tomografía computarizada de rayos X.; Permitir la obtención de imágenes desde estructuras subcelulares hasta organismos completos.

Principales métodos:

  • Desarrollo de una NanoPartícula de Selenio (cSeNP) conjugada con proteínas de aproximadamente 5 nm de diámetro.; Fusión del ADN que codifica la proteína cSeNP con el ADN que codifica FtsZ, un análogo de tubulina bacteriana.; Expresión de proteínas de fusión FtsZ-cSeNP en Escherichia coli y análisis posterior mediante tomografía electrónica de transmisión y microscopía de luz de fluorescencia.; Se utilizó tomografía computarizada de rayos X para evaluar el contraste en E. coli.

Principales resultados:

  • Los cSeNP exhiben un alto contraste en microscopía electrónica debido a su densidad electrónica.; Los experimentos confirmaron la localización de cSeNP correlacionada con las posiciones conocidas de los filamentos de FtsZ en E. coli.; Se encontró que menos del 5% de los cSeNP estaban en ubicaciones inesperadas.; La imagen de rayos X demostró un contraste distinguible del ruido de fondo.

Conclusiones:

  • El cSeNP sirve como un agente de contraste de imágenes versátil y clonable.; Facilita la localización y correlación de proteínas en todas las escalas de longitud biológica.; Esto es particularmente ventajoso para la microscopía electrónica, cerrando la brecha con técnicas de imagen de área más grande como la microscopía de fluorescencia.
