    Sus antecedentes:

    • La calorimetría isotérmica (IMC) mide directamente la producción de calor, lo que es crucial para comprender la termogénesis.
    • Los parámetros óptimos de IMC para la investigación del tejido adiposo aún no están definidos, lo que limita su aplicación.

    Objetivo del estudio:

    • Establecer y validar condiciones experimentales para cuantificar la termogénesis en el tejido adiposo mediante calorimetría isotérmica (IMC) de alto rendimiento.
    • Proporcionar pautas prácticas para el manejo de tejidos, el diseño de ensayos y la interpretación de datos.

    Principales métodos:

    • Se utilizó la plataforma de IMC de alto rendimiento CalScreener.
    • Se evaluó la termogénesis en organoides de adipocitos, adipocitos aislados y explantes de tejido adiposo.
    • Se investigó el impacto del manejo y almacenamiento de tejidos en la capacidad termogénica.

    Principales resultados:

    • La producción de calor en organoides se escaló con el tamaño y el número, mejorando la consistencia de la medición.
    • Los adipocitos aislados y los explantes intactos mostraron una termogénesis robusta y específica del depósito.
    • El tejido adiposo intacto mantuvo la capacidad termogénica ex vivo durante 6 horas en medio nutritivo.
    • Las condiciones de almacenamiento en frío, si bien preservaron la integridad mitocondrial, suprimieron la producción de calor basal.

    Conclusiones:

    • Se establecieron parámetros optimizados para la medición de la termogénesis del tejido adiposo mediante IMC.
    • Se demostró la viabilidad del análisis por lotes para cohortes grandes utilizando tejido adiposo ex vivo.
    • Se destacó la importancia del manejo y almacenamiento adecuados de los tejidos para evaluaciones precisas de la termogénesis.
