Principales métodos:
- Se utilizó un sistema de ratón transgénico inducible por Fos para expresar receptores de diseño quimiogenéticos activados exclusivamente por un fármaco de diseño (DREADDs) en neuronas activas. Se estimularon quimiogenéticamente conjuntos neuronales activados por anestesia para evaluar los efectos conductuales. Se empleó el aclaramiento de todo el cerebro iDisco+ y la microscopía de hoja de luz para mapear las neuronas capturadas.
Principales resultados:
- La estimulación de las neuronas activadas por anestesia causó inmovilidad, descoordinación y anti-nocicepción, aumentando la sensibilidad a la anestesia. La conciencia, evaluada por el reflejo de enderezamiento, no se perdió al estimular. Se revelaron neuronas escasas y distribuidas espacialmente con morfologías únicas en todo el cerebro.
Conclusiones:
- La anestesia general probablemente involucra circuitos neuronales específicos y distribuidos en lugar de actuar de manera inespecífica. Proporciona un mapa morfológico completo de todo el cerebro de las neuronas activadas por anestesia como un recurso para la investigación futura.
