These proteins can be classified into two general categories- proteins that generate mechanical forces and proteins that are subjected to mechanical forces.     Sus antecedentes:

    • La glicosilación de proteínas es crucial para las funciones celulares como el plegamiento y la estabilidad de las proteínas.
    • No se comprende bien el impacto de la glicosilación anormal en la función e interacciones de las proteínas.

    Objetivo del estudio:

    • Investigar sistemáticamente cómo la glicosilación afecta la estabilidad y la función de las proteínas.
    • Explorar las consecuencias de la glicosilación alterada en los procesos celulares y las redes de interacción de proteínas.

    Principales métodos:

    • Se utilizó proteómica basada en espectrometría de masas, glicobiología química y simulaciones de dinámica molecular.
    • Se alteró la vía secretora para generar proteínas con distintas estructuras de glicanos.
    • Se empleó el perfilado de proteoma térmico para analizar las consecuencias funcionales de los cambios en la glicosilación.

    Principales resultados:

    • Se observaron respuestas celulares al estrés convergentes, incluida la reorganización del tráfico y la redirección de proteínas hacia la degradación, tras las perturbaciones de la glicosilación.
    • Se identificaron modificaciones de glicanos terminales, específicamente la sialilación y la fucosilación, como las que tienen el impacto más significativo en la función de las proteínas de la superficie celular.
    • Se demostró que la pérdida de fucosilación en la integrina alfa 4 restringe la dinámica conformacional, reduciendo la afinidad de unión a VCAM-1.

    Conclusiones:

    • La regulación de la función proteica por la glicosilación es compleja y se extiende más allá de la simple identificación de glicoformas.
    • Las perturbaciones en la glicosilación terminal, particularmente la fucosilación, pueden alterar significativamente la función e interacciones de las proteínas.
    • Los hallazgos sugieren posibles estrategias terapéuticas dirigidas a la glicosilación para enfermedades como la esclerosis múltiple.
