Resumen

Este estudio presenta un flujo de trabajo novedoso para la síntesis y evaluación de quimeras dirigidas a la proteólisis (PROTAC). El enfoque directo a la biología acelera el descubrimiento de degradadores de proteínas dirigidos al eludir los pasos de purificación.

Sus antecedentes:

  • La degradación de proteínas dirigida es un campo en rápida evolución.
  • El desarrollo de quimeras dirigidas a la proteólisis (PROTAC) es un desafío debido a la falta de métodos de diseño racional.
  • Los métodos actuales a menudo implican procesos largos de síntesis y purificación.

Objetivo del estudio:

  • Desarrollar un flujo de trabajo optimizado para la síntesis y evaluación de PROTAC.
  • Superar las limitaciones del desarrollo tradicional de PROTAC.
  • Permitir la identificación y optimización rápidas de candidatos a fármacos PROTAC.

Principales métodos:

  • Un enfoque directo a la biología que combina la síntesis semi-automatizada de alto rendimiento con la evaluación directa de ensayos celulares.
  • Utilización de la cicloadición de azida-alquino catalizada por cobre para una síntesis eficiente de PROTAC a escala de nanomoles.
  • Implementación de un flujo de trabajo que evita el paso de purificación del compuesto.

Principales resultados:

  • Se demostró la aplicabilidad general del flujo de trabajo en cuatro objetivos diversos.
  • Se logró la síntesis de PROTAC en volúmenes de reacción tan bajos como 5 μL.
  • Se permitió la síntesis y evaluación de cientos de PROTAC en cuestión de días.

Conclusiones:

  • El flujo de trabajo desarrollado acelera significativamente el descubrimiento y desarrollo de PROTAC.
  • Este enfoque facilita la evaluación integral de la degradabilidad del objetivo y la selección de la ligasa E3.
  • La metodología ofrece una herramienta poderosa para avanzar en las estrategias de degradación de proteínas dirigidas.
