Sus antecedentes:

  • Las células epiteliales intestinales forman una barrera crítica, que regula las respuestas inmunitarias a través de la secreción asimétrica.; Comprender cómo los tipos de células epiteliales y los estímulos influyen en esta secreción es vital para la homeostasis intestinal.; Los modelos existentes carecen de la complejidad para recapitular completamente la fisiología intestinal humana in vivo.

Objetivo del estudio:

  • Establecer y caracterizar un modelo humano de organoide intestinal en chip (OoC).; Investigar la integridad de la barrera dependiente del tipo de célula y los perfiles de expresión génica.; Analizar el impacto de estímulos externos en el secretoma de las células epiteliales y la regulación inmunitaria.

Principales métodos:

  • Desarrollo y caracterización de un sistema humano de organoide intestinal en chip (OoC).; Secuenciación de ARN y microscopía para evaluar perfiles de expresión específicos de tipo celular.; Análisis del secretoma para evaluar patrones de secreción bidireccional en respuesta a diversos estímulos.

Principales resultados:

  • El modelo OoC recreó con éxito la función de barrera intestinal humana.; Demostró respuestas dependientes del tipo de célula a lipopolisacáridos (LPS), flagelina, gliadina y citoquinas.; Identificó patrones de secreción bidireccional asimétrica influenciados por estímulos específicos.

Conclusiones:

  • El modelo OoC desarrollado es una plataforma potente para estudiar la función de la barrera epitelial intestinal humana.; Este modelo proporciona información sobre la inmunidad innata mediada por células epiteliales en condiciones fisiológicas y fisiopatológicas.; El sistema OoC ofrece potencial para avanzar en estrategias terapéuticas personalizadas para enfermedades relacionadas con el intestino.
