La proteína 3 asociada al regulador del sustrato de la cinasa 5 dependiente de ciclina (CDK5RAP3) es crucial para la producción de testosterona en las células de Leydig. La reducción de los niveles de CDK5RAP3 se asocia con infertilidad masculina y alteración de la esteroidogénesis.

Sus antecedentes:

  • La producción de testosterona (T) por las células de Leydig (CL) es vital para la reproducción masculina, pero su regulación no se comprende completamente.
  • La proteína 3 asociada al regulador del sustrato de la cinasa 5 dependiente de ciclina (CDK5RAP3) se identificó como una proteína implicada en la esteroidogénesis y el metabolismo del colesterol.

Objetivo del estudio:

  • Investigar el papel de CDK5RAP3 en el desarrollo de las células de Leydig y la esteroidogénesis.
  • Explorar la posible relación entre la expresión de CDK5RAP3 y la infertilidad masculina.

Principales métodos:

  • Inmunoprecipitación-espectrometría de masas (IP-MS) en tejido testicular de ratón.
  • Análisis de la expresión de CDK5RAP3 en muestras humanas y en un modelo de rata inducido por etano dimetanosulfonato (EDS).
  • Estudios funcionales en CL primarias y células MLTC-1, incluyendo silenciamiento y sobreexpresión de CDK5RAP3.
  • Estudios in vivo utilizando silenciamiento de CDK5RAP3 mediado por AAV2/9 en ratones adultos.
  • Estudios mecanísticos que involucran interacciones proteína-proteína y análisis de vías (señalización BMP).

Principales resultados:

  • La expresión de CDK5RAP3 se redujo en las CL de pacientes con fallo espermatogénico y aumentó durante la maduración y regeneración de las células de Leydig.
  • La translocación nuclear de CDK5RAP3 ocurrió tras la estimulación con hCG; su silenciamiento alteró la producción de testosterona y redujo la expresión de genes esteroidogénicos (STAR, CYP11A1, CYP17A1, HSD3B).
  • La sobreexpresión de CDK5RAP3 mejoró la producción de testosterona; el silenciamiento in vivo redujo significativamente los niveles séricos de testosterona.
  • CDK5RAP3 interactuó con SMAD4 y CEBPB, y la inhibición de la vía BMP rescató los déficits de testosterona.

Conclusiones:

  • CDK5RAP3 es un regulador esencial de la esteroidogénesis de las células de Leydig.
  • CDK5RAP3 desempeña un papel importante en el mantenimiento de los niveles de testosterona y puede ser relevante para la infertilidad masculina asociada con bajos niveles de testosterona.
