Resumen

Los niveles séricos de N-Acetil-D-manosamina (ManNAc) y ácido N-acetilneuramínico (Neu5Ac) varían según la raza, el sexo, la edad y el IMC. Este estudio desarrolló un método LC-MS/MS preciso para cuantificar estos componentes de glicosilación.

Sus antecedentes:

  • La N-Acetil-D-manosamina (ManNAc) y el ácido N-acetilneuramínico (Neu5Ac) son cruciales para la glicosilación y las funciones fisiológicas.
  • La influencia de factores demográficos como la edad, el IMC, la raza y el sexo en los niveles séricos de ManNAc y Neu5Ac sigue estando en gran medida sin caracterizar.

Objetivo del estudio:

  • Desarrollar y validar un método sensible de cromatografía líquida-espectrometría de masas en tándem (LC-MS/MS) para la cuantificación simultánea de ManNAc y Neu5Ac en suero humano.
  • Investigar las asociaciones entre los niveles séricos de ManNAc y Neu5Ac y los factores demográficos (edad, IMC, raza, sexo) en una cohorte humana.

Principales métodos:

  • Desarrollo y validación de un ensayo LC-MS/MS simultáneo para ManNAc y Neu5Ac séricos.
  • Análisis de 155 muestras de suero de la cohorte Adventist Health Study 2 (AHS-2).
  • Análisis estadístico para determinar las correlaciones entre los niveles de analitos y las variables demográficas.

Principales resultados:

  • Los niveles séricos de Neu5Ac fueron significativamente más altos en mujeres (p=0,029 para conjugados, p=0,026 para totales).
  • Los niveles de ManNAc conjugado y total fueron significativamente más altos en participantes negros en comparación con participantes blancos (p < 0,001).
  • Un IMC más alto se asoció con un aumento de ManNAc libre (p=0,041), y una edad más avanzada se asoció con ManNAc libre y total más altos (p ≤ 0,001 y p=0,045, respectivamente).

Conclusiones:

  • El método LC-MS/MS validado proporciona una cuantificación sensible y específica de ManNAc y Neu5Ac séricos.
  • Las concentraciones séricas de ManNAc y Neu5Ac están influenciadas por la raza, el sexo, la edad y el IMC.
  • Esta metodología permitirá una mayor investigación sobre los roles fisiológicos y la relevancia clínica de estos componentes de glicosilación.
