Resumen

Objetivo del estudio:

  • Investigar los efectos inmunomoduladores de los lisados de cuatro especies bacterianas: Bacteroides vulgatus (BV), Clostridium perfringens (CP), Parabacteroides distasonis (PD) y Ruminococcus albus (RA).
  • Determinar el impacto de estos lisados bacterianos en la secreción de IL-17, INF-γ, IL-2 y TGF-β1 por parte de células humanas HT-29, células mononucleares de sangre periférica (PBMC) y monocitos (MON).
  • Evaluar las respuestas dependientes de la dosis de las células inmunitarias a los lisados bacterianos.

Principales métodos:

  • Incubación de células humanas HT-29, PBMC y MON con diferentes dosis (100 µg y 400 µg) de lisados bacterianos (BV, CP, PD, RA).
  • Medición de la secreción de citoquinas (IL-17, INF-γ, IL-2, TGF-β1) mediante ensayos apropiados.
  • Análisis estadístico para comparar los niveles de citoquinas entre los grupos tratados y los controles.

Principales resultados:

  • Los lisados de CP, RA y PD redujeron significativamente la secreción de IL-17 por parte de las células HT-29 de manera dependiente de la dosis.
  • Los lisados de BV y PD disminuyeron significativamente la secreción de IL-17 por parte de las células mononucleares de sangre periférica (PBMC) y los monocitos (MON).
  • Los lisados de PD y CP redujeron la secreción de INF-γ por parte de las células HT-29, mientras que los lisados de CP y BV redujeron el INF-γ en MON y PBMC, respectivamente.

Conclusiones:

  • Los lisados bacterianos seleccionados, en particular CP y PD, pueden reducir significativamente la secreción de IL-17 e INF-γ por parte de las células inmunitarias humanas (HT-29, PBMC, MON).
  • Los efectos inmunomoduladores observados dependen de la dosis.
  • La magnitud de la reducción de citoquinas en este estudio sugiere un impacto limitado en el estado inmunitario sistémico general.
