Resumen

Los hidrogeles de complejos polielectrolíticos (PEC) de tribloques ofrecen una entrega de proteínas sintonizable. Los aditivos proteicos no alteran la microestructura del hidrogel PEC, lo que permite un control preciso sobre el hinchamiento y la liberación para aplicaciones de administración de fármacos.

Sus antecedentes:

  • Los hidrogeles de complejos polielectrolíticos (PEC) de tribloques son materiales autoensamblados con propiedades sintonizables, adecuados para la entrega de proteínas.
  • Comprender el impacto de los aditivos proteicos en la microestructura y el hinchamiento de los hidrogeles PEC es crucial para su aplicación como vehículos de entrega.

Objetivo del estudio:

  • Investigar la influencia de las proteínas modelo en la microestructura y el comportamiento de hinchamiento de los hidrogeles PEC.
  • Demostrar el control preciso sobre las características de hinchamiento, disolución y liberación de proteínas de los hidrogeles PEC.
  • Establecer parámetros de diseño para la liberación de proteínas sintonizable a partir de hidrogeles PEC.

Principales métodos:

  • Dispersión de rayos X en ángulo pequeño (SAXS) para analizar la microestructura del hidrogel.
  • Caracterización de las propiedades de hinchamiento y disolución del hidrogel.
  • Evaluación de la cinética de liberación de proteínas en condiciones de composiciones variables del hidrogel.

Principales resultados:

  • Las proteínas modelo (BSA, lipasa, CAII, ureasa) no alteraron la microestructura del hidrogel PEC a concentraciones relevantes.
  • La longitud del extremo del bloque polielectrolítico de tribloque (tbPE) y la concentración controlaron con precisión el hinchamiento y la disolución.
  • Las propiedades del hidrogel PEC (hinchamiento, disolución, módulos de cizallamiento) no se vieron afectadas por la inclusión de proteínas.
  • Se logró una liberación de proteínas sintonizable variando la concentración de tbPE, la longitud del extremo del bloque, la mezcla de tbPE y la incorporación de redes interpenetrantes.

Conclusiones:

  • Los hidrogeles PEC son plataformas accesibles para la entrega de proteínas, no afectadas por aditivos proteicos.
  • Los parámetros de diseño accesibles permiten una sintonización precisa de las características de hinchamiento y liberación de proteínas.
  • Este estudio proporciona información sobre las interacciones macromoleculares dentro de los autoensamblajes basados en PEC para la administración de fármacos.
