Las células tumorales interactúan con su microambiente, impulsando paradójicamente el crecimiento del cáncer. Comprender estas complejas redes celulares es clave para desarrollar nuevas terapias que mejoren la inmunidad antitumoral y superen la resistencia al tratamiento.

Sus antecedentes:

  • El microambiente tumoral (TME) comprende células malignas, estromales, inmunitarias y endoteliales que interactúan mediante contacto directo o factores solubles.
  • La composición tumoral y la infiltración de células inmunitarias evolucionan, creando nichos dinámicos y heterogéneos.
  • Estas interacciones celulares dentro del TME influyen en la defensa inmunitaria y la progresión tumoral.

Objetivo del estudio:

  • Integrar el conocimiento sobre las interacciones celulares dentro del TME.
  • Comprender cómo estas redes apoyan paradójicamente la progresión tumoral al tiempo que involucran a las células inmunitarias.
  • Identificar estrategias terapéuticas dirigidas a las redes de comunicación pro-tumoral.

Principales métodos:

  • Revisión de la literatura e integración de conocimientos.
  • Análisis de la participación de las células inmunitarias y los procesos homeostáticos mediados por células estromales.
  • Exploración de la reprogramación metabólica, el estrés oxidativo y la señalización por vesículas extracelulares.

Principales resultados:

  • Las interacciones celulares en el TME son complejas y dinámicas, influyendo en el desarrollo tumoral.
  • Las células inmunitarias y los componentes estromales desempeñan roles paradójicos, promoviendo a veces el crecimiento tumoral.
  • Los cambios metabólicos, el estrés oxidativo y las vesículas extracelulares contribuyen a la resistencia tumoral.

Conclusiones:

  • Se necesita un marco integral para comprender las interacciones celulares del TME.
  • La modulación de las redes de comunicación pro-tumoral puede mejorar la inmunidad antitumoral.
  • El desarrollo de inmunoterapias multiobjetivo es crucial para controlar el cáncer y superar la resistencia.
