Sus antecedentes:

  • La patogénesis de la enfermedad pulmonar intersticial relacionada con la artritis reumatoide (DI-AR) sigue siendo poco comprendida.
  • Aclarar las diferencias celulares y transcriptómicas en los pulmones con DI-AR es crucial para comprender los mecanismos de la enfermedad.

Objetivo del estudio:

  • Elucidar el panorama celular y transcriptómico de las células epiteliales e inmunitarias en los pulmones con DI-AR.
  • Comparar las células Tph pulmonares en la DI-AR con las células Tph sinoviales.

Principales métodos:

  • Secuenciación de ARN unicelular de explantes pulmonares de pacientes con DI-AR, controles y pacientes con DI-CTD.
  • Análisis integrador de subagrupaciones de células T con datos públicos de células T sinoviales.
  • Tinción de inmunofluorescencia de secciones pulmonares para la validación celular.

Principales resultados:

  • Identificadas 18 agrupaciones celulares distintas a partir de 184.814 células perfiladas.
  • Observado una reducción de las células alveolares tipo 2 y un aumento de las células basales/ciliadas en los pulmones con DI-AR.
  • Descubierta la presencia exclusiva de células T periféricas colaboradoras (Tph) en los pulmones con DI-AR, con una activación distinta y firmas citotóxicas/agotadas más bajas en comparación con las células Tph sinoviales.

Conclusiones:

  • El análisis transcriptómico detallado revela diferencias celulares clave en los pulmones con DI-AR.
  • Las células T periféricas colaboradoras (Tph) son un sello distintivo de los pulmones con DI-AR.
  • La comparación entre tejidos resalta las características específicas de los órganos de las células Tph.
