Rab GTPases act in a regulated cascade during membrane fusion, helping the lipid bilayers mix. The Rab family of proteins are active when bound to GTP, and inactive when bound to GDP. Hence, they act as guanine nucleotide-dependent molecular switches. Rab-GTP recognizes and binds to long or short-range tethering proteins to capture the target vesicle. These tethers coordinate with SNAREs on the vesicle and the target membrane to assemble the trans SNARE complex that locks the mixing bilayers.

While every living organism has a genome of some kind (be it RNA, or DNA), there is considerable variation in the sizes of these blueprints. One major factor that impacts genome size is whether the organism is prokaryotic or eukaryotic. In prokaryotes, the genome contains little to no non-coding sequence, such that genes are tightly clustered in groups or operons sequentially along the chromosome. Conversely, the genes in eukaryotes are punctuated by long stretches of non-coding sequence.

Sus antecedentes:

  • El modelo de cascada inhibitoria (ICM) vincula el desarrollo con la variación evolutiva.
  • Propone que los rasgos seriales como los dientes molares son controlados por procesos de activación e inhibición.
  • Las aplicaciones anteriores muestran un buen ajuste tanto para rasgos secuenciales como no secuenciales.

Objetivo del estudio:

  • Reevaluar críticamente el modelo de cascada inhibitoria (ICM).
  • Investigar si el ajuste del ICM es un artefacto de la estandarización estadística.
  • Probar la validez del ICM en los niveles de desarrollo, variacional y evolutivo.

Principales métodos:

  • Se rearticuló la base matemática y conceptual del ICM.
  • Se aplicó el ICM rearticulado a datos biológicos, no biológicos y simulados.
  • Se analizaron los datos utilizando métodos estadísticos para evaluar el ajuste del modelo.

Principales resultados:

  • Se demostró que el ajuste aparente del bondad del modelo ICM es un artefacto de la escala de valores correlacionados.
  • No se encontró evidencia que respalde el ICM a nivel de desarrollo.
  • No se encontró evidencia que respalde el ICM a nivel variacional o evolutivo.

Conclusiones:

  • El modelo de cascada inhibitoria (ICM) no proporciona una explicación válida para la morfogénesis.
  • El éxito percibido del modelo es un artefacto de los métodos estadísticos, no de los procesos biológicos.
  • No existe apoyo empírico para el ICM en el desarrollo, la variación o la evolución.
