Resumen

Los dendrímeros diseñados entregan efectivamente diclofenaco, un medicamento antiinflamatorio, como una terapia contra el cáncer. Los nanocarriers muestran una mejor matanza de las células cancerosas y una toxicidad reducida para las células sanas, ofreciendo una prometedora estrategia de reutilización de fármacos.

Sus antecedentes:

  • El desarrollo de fármacos contra el cáncer enfrenta altos costos y un éxito limitado, lo que requiere la reutilización de fármacos.
  • El diclofenaco, un AINE, muestra un potencial anticancerígeno, pero tiene una mala solubilidad y un aclaramiento rápido.
  • Se necesitan nuevos nanocarriers para mejorar la idoneidad quimioterapéutica del diclofenaco.

Objetivo del estudio:

  • Diseñar dendrímeros de poliéster heterofuncionales PEGilados (HFD) para la administración controlada de diclofenaco.
  • Evaluar la eficacia contra el cáncer y la selectividad de los HFD cargados de diclofenaco.
  • Para investigar el mecanismo de acción, incluida la generación de especies reactivas de oxígeno (ROS).

Principales métodos:

  • La conjugación de diclofenaco a través de la cicloadición azida-alquina catalizada por cobre (CuAAC).
  • PEGilación periférica a través de la esterificación del anhídrido.
  • Caracterización de las construcciones dendrímeras G1 y G2 (G1- ((Dicl) 3- ((mPEG) 6 y G2- ((Dicl) 9- ((mPEG) 12).
  • Evaluación in vitro de la citotoxicidad en líneas celulares de cáncer y fibroblastos.
  • Evaluación del nivel de ROS y estudios mecanicistas.

Principales resultados:

  • Se formaron nanoestructuras anfifílicas de núcleo y cáscara con diámetros hidrodinámicos de 170-330 nm.
  • G1-(Dicl) 3-(mPEG) 6 mostró una reducción significativa de la viabilidad de las células cancerosas (50-70%) a 1-10 μM con alta selectividad contra los fibroblastos (>20 veces mejor índice terapéutico).
  • Ambos dendrímeros indujeron ROS en concentraciones significativamente más bajas que el diclofenaco libre, en correlación con la citotoxicidad.
  • G2- ((Dicl) 9- ((mPEG) 12 demostró una actividad potente pero dependiente de la línea celular.

Conclusiones:

  • Los HFD son una versátil plataforma de nanomedicina para reutilizar medicamentos como el diclofenaco.
  • El dendrimero G1 ofrece un equilibrio óptimo de eficacia, selectividad y potencial de traducción para la terapia del cáncer.
  • La citotoxicidad mediada por ROS es un mecanismo clave para los dendrímeros cargados de diclofenaco.
