Desarrollamos una nueva herramienta de diagnóstico basada en CRISPR que vincula los eventos de reparación del ADN con la activación de CRISPR-Cas12a. Este sistema permite la detección sensible de la actividad de la enzima de reparación del ADN y facilita la detección de fármacos.

Objetivo del estudio:

  • Desarrollar una plataforma sintética que acople la actividad de reparación del ADN con la activación de CRISPR-Cas12a.
  • Para crear un método para detectar la actividad de la enzima de reparación del ADN y permitir la detección de drogas.

Principales métodos:

  • Desarrolló una plataforma de transducción sintética que vincula los eventos de reparación por escisión de la base (BER) a la activación de CRISPR-Cas12a.
  • Utilizó glicosilasas de ADN (UDG, hOGG1) y un transductor de ADN programable.
  • Convirtió la actividad enzimática en una señal de fluorescencia a través de la escisión colateral Cas12a.

Principales resultados:

  • Logró una detección rápida, sensible y específica basada en lisato de la actividad de reparación del ADN.
  • Adaptabilidad demostrada para el cribado de alto rendimiento de inhibidores de moléculas pequeñas.
  • Estableció un marco general para la transducción de la reparación del ADN en salida CRISPR programable.

Conclusiones:

  • La plataforma sintética permite el acoplamiento directo de la reparación del ADN a CRISPR-Cas12a.
  • Este enfoque facilita la detección bioanalítica sensible y el cribado de drogas.
  • Abre el camino para los circuitos de genes sintéticos sensibles a la reparación del ADN y el descubrimiento de fármacos basados en CRISPR.
