Resumen
El diseño de inmunoterápicos nanoestructurados como los ácidos nucleicos esféricos (SNA) es clave para la terapia del cáncer. Una variante específica de SNA, N-HSNA, estimuló significativamente las respuestas inmunes y redujo el crecimiento tumoral en modelos preclínicos.

Principales resultados:
- Todos los SNAs aumentaron la activación de las células inmunes y la citotoxicidad en comparación con una mezcla simple de péptido-CpG.
- N-HSNA demostró un rendimiento superior, induciendo una secreción de interferón-γ significativamente mayor y una citotoxicidad de las células T CD8 +.
- In vivo, el N-HSNA redujo la carga tumoral, prolongó la supervivencia y expandió las células T CD8 +.
- El análisis transcriptómico reveló que N-HSNA promovió la activación inmune y redujo los marcadores de agotamiento de las células T.
- N-HSNA aumentó significativamente la citotoxicidad contra los esferoides de cáncer de cabeza y cuello derivados del VPH+ derivados de pacientes.
Conclusiones:
- La colocación y orientación del antígeno dentro de los SNA son parámetros críticos para maximizar la potencia inmunoterapéutica.
- N-HSNA representa un prometedor candidato inmunoterapéutico nanoestructurado para los cánceres asociados al VPH.
