Los microplásticos de la dieta (MP) alteran la salud intestinal, causando estrés oxidativo e inflamación. El envejecimiento ambiental de los MP puede alterar su toxicidad, afectando la homeostasis intestinal y la composición de la microbiota.

Sus antecedentes:

  • Los microplásticos dietéticos (MP) son cada vez más reconocidos como contaminantes ambientales con posibles impactos en la salud.
  • Los estudios anteriores a menudo usaban partículas no envejecidas, que no reflejan la exposición y la toxicidad de MP en el mundo real.
  • El envejecimiento ambiental puede alterar las propiedades físicas y químicas de los MP, influyendo potencialmente en sus efectos biológicos.

Objetivo del estudio:

  • Para investigar la toxicidad intestinal in vivo de microplásticos de PVC envejecidos y no envejecidos después de una exposición oral subcrónica.
  • Evaluar el impacto de los MP de PVC en el estrés oxidativo, la inflamación, la microbiota intestinal y la histología intestinal.
  • Para determinar si el envejecimiento de las partículas influye en la toxicidad de los microplásticos en el intestino.

Principales métodos:

  • Se administró por vía oral a ratones hembras BALB/c MPs de PVC envejecidos o no envejecidos (0,02 mg/día) durante 70 días.
  • Marcadores de estrés oxidativo evaluados (ROS / RNS, peroxidación lipídica), citoquinas inflamatorias (IL-1β, TNF-α), cambios histológicos, microbiota intestinal y albúmina fecal.
  • Acumulación cuantificada de MP mediante digestión asistida por microondas y espectroscopia μRaman.

Principales resultados:

  • Tanto los MP de PVC envejecidos como los no envejecidos indujeron un estrés oxidativo significativo y inflamación en el intestino.
  • El análisis histológico reveló una reducción de la profundidad de la cripta y un aumento de las células secretoras de moco.
  • La exposición a MP alteró la composición de la microbiota intestinal, redujo los niveles de ácidos grasos de cadena corta (SCFA) y mostró diferencias en la toxicidad entre las partículas envejecidas y no envejecidas.

Conclusiones:

  • La exposición subcrónica a los microplásticos de PVC interrumpe la homeostasis intestinal a través del estrés oxidativo, la inflamación y la disbiosis intestinal.
  • El proceso de envejecimiento de los microplásticos puede modular su toxicidad intestinal.
  • Estos hallazgos resaltan la importancia de considerar el envejecimiento ambiental al evaluar los riesgos de los microplásticos para la salud intestinal.
