Sus antecedentes:

  • El foto blanqueamiento limita las imágenes a largo plazo de células vivas de procesos biológicos dinámicos.
  • Los luminógenos de emisión inducida por agregación polimérica (AIEgens) ofrecen fotostabilidad, pero a menudo se localizan en los lisosomas.
  • Existe la necesidad de sondas con una mayor accesibilidad subcelular y una mejor fotoestabilidad.

Objetivo del estudio:

  • Desarrollar un nuevo enfoque para sintetizar sondas fluorescentes poliméricas fotostables in situ dentro de orgánulos específicos.
  • Para superar las limitaciones de las sondas actuales, en particular la localización lisosómica y el fotoblanqueamiento.
  • Para permitir imágenes a largo plazo en tiempo real de la dinámica celular, específicamente las funciones mitocondriales.

Principales métodos:

  • Se empleó la estrategia de polimerización bioortogonal inducida por localización orgánica (OLIBOP, por sus siglas en inglés).
  • Se diseñó un precursor de moléculas pequeñas (1-AIE) con propiedades AIE, porción dirigida a las mitocondrias y un grupo de condensación bioortogonal.
  • La localización mitocondrial y la polimerización in situ se desencadenaron por un enlace disulfuro sensible a GSH, visualizado con el uso de phasor-FLIM.

Principales resultados:

  • El precursor diseñado (1-AIE) se polimerizó con éxito in situ específicamente en los sitios mitocondriales.
  • El resultante poli-AIEgen formado in situ exhibió una intensidad de fluorescencia significativamente mejorada y una vida útil de fluorescencia más larga.
  • La fotostabilidad excepcional permitió un seguimiento en tiempo real sin precedentes de la dinámica mitocondrial durante períodos prolongados.

Conclusiones:

  • OLIBOP proporciona un enfoque innovador para superar el compromiso entre la entrega de la sonda y la estabilidad.
  • Este método permite la creación de sondas poliméricas fotostables dentro de los orgánulos objetivo, mejorando la biocompatibilidad.
  • Las sondas desarrolladas facilitan la obtención de imágenes de células vivas a largo plazo de alta resolución de los procesos celulares, particularmente la dinámica mitocondrial.
