Resumen
La focalización del factor de transcripción EB (TFEB) en microglia mejora la eliminación de beta-amiloide y reduce la neuroinflamación, mejorando la función cognitiva en modelos de enfermedad de Alzheimer.

Sus antecedentes:
- La enfermedad de Alzheimer (EA) implica la acumulación de beta-amiloide (Aβ) y neuroinflamación.
- La disfunción de la vía autofágica-lisosomal (ALP) en microglia es un factor clave en la progresión de la EA.
- El factor de transcripción EB (TFEB) regula la ALP, pero su papel en microglia en la EA no está claro.
Objetivo del estudio:
- Investigar el potencial terapéutico de la sobreexpresión de TFEB en microglia en la patogénesis de la enfermedad de Alzheimer (EA).
- Determinar si la mejora de TFEB en microglia puede mitigar la patología relacionada con la EA y los déficits cognitivos.
Principales métodos:
- Desarrollo de un modelo de ratón 5xFAD con sobreexpresión de TFEB específica de microglia (5xTFEB).
- Realización de pruebas conductuales, histopatología, análisis bioquímicos e imagenología de células vivas de la fagocitosis de Aβ.
- Utilización de secuenciación de ARN masiva para análisis de expresión génica diferencial y evaluación de la activación de la inflamación.
Principales resultados:
- La sobreexpresión de TFEB en microglia restauró la función de la vía autofágica-lisosomal (ALP) y promovió la eliminación fagolisosomal de Aβ.
- Redujo la carga amiloide en regiones cerebrales clave y rescató los déficits de memoria en ratones 5xTFEB.
- El análisis transcriptómico mostró una ALP regulada al alza y una señalización inflamatoria regulada a la baja, con una atenuación de la activación de la inflamación.
Conclusiones:
- La activación dirigida de TFEB en microglia mejora la eliminación de Aβ y alivia la neuroinflamación y el deterioro cognitivo en la EA.
- La modulación de TFEB en microglia representa una estrategia terapéutica prometedora específica de tipo celular para la EA.
- Este enfoque también puede beneficiar a otros trastornos neurodegenerativos caracterizados por disfunción autofágica e inflamación.
