Resumen
Un nuevo anticuerpo derivado de tiburón (VNAR) se dirige selectivamente al receptor MET para posibles aplicaciones teranósticas en el cáncer de pulmón no microcítico (CPNM). Este agente muestra promesa tanto para la obtención de imágenes como para el tratamiento del CPNM alterado por MET.

Sus antecedentes:
- Las alteraciones del receptor de tirosina quinasa MET ocurren en aproximadamente el 6% de los cánceres de pulmón no microcíticos (CPNM).
- La sobreexpresión de MET también se observa en el CPNM, lo que resalta su papel en la progresión del cáncer.
Objetivo del estudio:
- Desarrollar un nuevo receptor de antígeno variable de dominio único (VNAR) derivado de tiburón para aplicaciones teranósticas dirigidas al receptor MET.
- Evaluar la eficacia del agente basado en VNAR tanto para la obtención de imágenes como para la intervención terapéutica en el CPNM alterado por MET.

- Marcaje con radioisótopos de zirconio-89 ([ 89 Zr]Zr-vMET1-Fc) para la obtención de imágenes por PET/CT.
- Marcaje con radioisótopos de lutecio-177 ([ 177 Lu]Lu-vMET1-Fc) para la evaluación terapéutica.
- Evaluación de la biodistribución y seguridad en estudios con primates no humanos.
Principales resultados:
- Un clon de VNAR con alta selectividad por MET fue identificado.
- vMET1-Fc demostró internalización selectiva en líneas celulares y xenoinjertos que expresan MET.
- [ 89 Zr]Zr-vMET1-Fc permitió la detección por PET/CT de xenoinjertos de CPNM positivos para MET.
- [ 177 Lu]Lu-vMET1-Fc retrasó el crecimiento tumoral en xenoinjertos mutantes para MET y amplificados para MET.
- Los estudios en primates no humanos mostraron una biodistribución favorable, una eliminación predecible y una captación mínima fuera del objetivo.
