Resumen

Los investigadores desarrollaron una nueva secuencia de direccionamiento mitocondrial (RMTS) para administrar herramientas de edición génica CRISPR en las mitocondrias. Este avance permite la corrección eficiente de los defectos del ADN mitocondrial (ADNmt), ofreciendo una nueva esperanza para el tratamiento de las enfermedades genéticas mitocondriales.

Sus antecedentes:

  • Las enfermedades genéticas mitocondriales son causadas por defectos en el ADN mitocondrial (ADNmt).
  • Los enfoques actuales de terapia génica enfrentan desafíos en la entrega de componentes de CRISPR, específicamente ARN guía únicos (sgRNA), en las mitocondrias.
  • Los sistemas de entrega eficientes son cruciales para el desarrollo de terapias efectivas mediadas por CRISPR del ADNmt.

Objetivo del estudio:

  • Identificar y diseñar una secuencia de direccionamiento mitocondrial para la entrega eficaz de componentes de CRISPR-Cas9 en las mitocondrias.
  • Desarrollar una nueva herramienta de terapia génica para corregir mutaciones del ADNmt.
  • Evaluar la eficacia de la herramienta diseñada en modelos celulares y in vivo.

Principales métodos:

  • Análisis de redes de ARN largos no codificantes (lncRNAs) y proteínas de unión a ARN (RBPs) localizados en mitocondrias para identificar posibles mecanismos de direccionamiento.
  • Derivación y optimización de una secuencia de direccionamiento mitocondrial de 30 nucleótidos (ST2-RNA RMTS) del lncRNA RP11-46H11.3.
  • Ingeniería de la herramienta RMTS-CRISPR mediante la fusión del ST2-RMTS al sgRNA.
  • Validación de la eficiencia de direccionamiento y escisión del ADNmt en modelos celulares y in vivo.

Principales resultados:

  • Identificación del lncRNA RP11-46H11.3 como portador de translocación mitocondrial a través de RBP.
  • El ST2-RMTS derivado demostró una alta eficiencia de localización mitocondrial.
  • La herramienta RMTS-CRISPR de ingeniería se dirigió y escindió con éxito el ADNmt.
  • Se lograron eficiencias significativas de cambio de heteroplasmia de ADNmt de hasta el 26,37 % en modelos celulares y del 26,79 % in vivo para la mutación m.3243A>G.

Conclusiones:

  • Se ha desarrollado una nueva estrategia de intervención génica mitocondrial basada en CRISPR utilizando un RMTS diseñado.
  • La herramienta RMTS-CRISPR ofrece un enfoque tecnológico prometedor para corregir mutaciones heteroplásmicas del ADNmt.
  • Esta estrategia tiene aplicaciones potenciales para el tratamiento de diversos trastornos mitocondriales.
