Resumen

Las características radiómicas altamente estables, basadas principalmente en la textura de regiones tumorales enteras, mejoran los modelos de caracterización de tumores cerebrales. Sus antecedentes:

  • La generalizabilidad de los modelos radiogenómicos es crucial para su aplicación clínica.

Objetivo del estudio:

  • Para evaluar la estabilidad de las características radiómicas derivadas de máscaras de segmentación profunda totalmente automáticas en tumores cerebrales.
  • Para evaluar el impacto de la estabilidad de las características en las tareas de predicción aguas abajo, específicamente la predicción de mutaciones IDH.
  • Determinar si las características radiómicas estables y discriminatorias pueden conducir a modelos radiogenómicos generalizables.

Principales métodos:

  • Utilizó el conjunto de datos BraTS 2020 para la segmentación de resonancia magnética 3D multiparamétrica y la predicción de IDH.
  • Empleó siete redes neuronales convolucionales (CNN) de última generación para la segmentación tumoral multirregional totalmente automática.
  • Se evaluó la estabilidad de las características radiómicas utilizando el Coeficiente de Correlación de Concordancia General (OCCC) y las características discriminatorias seleccionadas a través de RFE-SVM.

Principales resultados:

  • Las características radiómicas altamente estables se basaron predominantemente en la textura (79,1%), principalmente de toda la región del tumor (WT) (96,1%).
  • La estabilidad de la característica fue más alta para WT (OCCC: 0.87 ± 0.12), seguida por el núcleo tumoral (TC) y el núcleo de mejora (EC).
  • El filtrado de estabilidad mejoró significativamente el rendimiento predictivo (AUC: 0.81 ± 0.02 a 0.94 ± 0.006) y redujo la variabilidad (RSD: 2.28% a 0.64%).

Conclusiones:

  • Se pueden desarrollar modelos radiogenómicos robustos y generalizables utilizando características radiómicas estables y discriminatorias.
  • La estabilidad de las características es un factor crítico para la caracterización y predicción confiables de tumores cerebrales.
  • Los hallazgos apoyan el uso de características radiómicas estables para construir herramientas de diagnóstico basadas en IA confiables.
