Sus antecedentes:
- Las células madre de la pulpa dental (DPSCs) tienen limitaciones para uso clínico debido a bajo rendimiento y capacidad reducida en etapas posteriores.
- La endodoncia regenerativa busca restaurar el complejo dentina-pulpa, incluida la inervación, yendo más allá de la simple revascularización.
- Los andamios de pulpa dental descelularizada (dDP) ofrecen una plataforma prometedora para la ingeniería de tejidos.
Objetivo del estudio:
- Investigar las células madre mesenquimales del cordón umbilical (WJ-MSC) como fuente de células alogénicas para la regeneración pulpar.
- Evaluar la combinación de WJ-MSC con andamios dDP para la regeneración de tejidos funcionales.
- Evaluar el potencial de diferenciación y las respuestas biológicas de las WJ-MSC en andamios dDP.
Principales métodos:
- Descelularización de tejidos de pulpa dental humana y caracterización de andamios.
- Repoblación de andamios dDP con WJ-MSC.
- Evaluación de la adhesión, migración, diferenciación osteogénica, odontogénica y neurogénica de WJ-MSC.
- Análisis de expresión génica de marcadores de diferenciación y regulatorios.
Principales resultados:
- Decelularización exitosa de andamios dDP, preservando la estructura de la matriz extracelular.
- Las WJ-MSC se adhirieron y migraron dentro de los andamios dDP.
- Los andamios repoblados promovieron la diferenciación osteogénica y odontogénica de las WJ-MSC.
- Se observó una regulación al alza del marcador neurogénico NCAM y una regulación a la baja de marcadores inhibidores, angiogénicos y reguladores inmunes.
Conclusiones:
- La combinación de WJ-MSC con andamios dDP es un enfoque factible para la regeneración pulpar funcional.
- Esta estrategia presenta una vía prometedora para la traducción clínica en endodoncia regenerativa.
- Las WJ-MSC en andamios dDP demuestran potencial para restaurar la vitalidad y función pulpar.
