Sus antecedentes:

  • Los anticuerpos monoclonales de inmunoglobulina G4 (IgG4 mAbs) son terapias cruciales pero poseen vulnerabilidades estructurales únicas. Los surfactantes no iónicos como los polisorbatos (Tween 20/80) y el poloxámero 188 (P188) se usan comúnmente para estabilizar formulaciones de proteínas. Comprender los mecanismos por los cuales estos surfactantes protegen las IgG4 mAbs bajo diversas condiciones de estrés es esencial para el diseño racional de formulaciones.

Objetivo del estudio:

  • Elucidar los mecanismos de estabilización dependientes del estrés de los polisorbatos y el poloxámero 188 para las IgG4 mAbs. Investigar el papel de las interacciones proteína-surfactante, la concentración micelar crítica (CMC) y la cinética de adsorción en la estabilización. Establecer un marco que conecte las interacciones fundamentales con las estrategias de formulación para las IgG4 mAbs.

Principales métodos:

  • Se utilizó calorimetría de titulación isotérmica (ITC) para estudiar la termodinámica de la unión proteína-surfactante. Se midió la concentración micelar crítica (CMC) para comprender el comportamiento de agregación del surfactante. Se empleó un modelo de estrés múltiple que incluye agitación orbital, estrés de vórtice y ciclos de congelación-descongelación para evaluar la eficacia de la estabilización. Se investigaron los mecanismos de estabilización en condiciones de estrés tanto termodinámicas como cinéticas controladas.

Principales resultados:

  • Los polisorbatos interactúan con IgG4 a través de interacciones hidrofóbicas impulsadas por la entropía, mientras que P188 no muestra una unión significativa. Ambos surfactantes estabilizan IgG4 a concentraciones superiores a su CMC mediante adsorción interfacial bajo estrés leve. Bajo un estrés intenso de vórtice, los polisorbatos requieren concentraciones supra-CMC para una protección completa, actuando como reservorios de monómeros, mientras que P188 ofrece una estabilización limitada debido a una cinética de adsorción más lenta. Los mecanismos de estabilización son distintos y dependen del tipo e intensidad del estrés aplicado.

Conclusiones:

  • La estabilización de IgG4 por surfactantes no iónicos se rige por vías distintas y dependientes del estrés. Los polisorbatos y P188 exhiben diferentes mecanismos de estabilización basados en sus perfiles de interacción y cinética de adsorción. Los hallazgos facilitan un cambio de la selección empírica a la selección guiada por mecanismos de estabilizadores para formulaciones de IgG4 mAb, mejorando la estabilidad y eficacia del producto farmacéutico.
