Sus antecedentes:

  • La tractografía por resonancia magnética de difusión (dMRI) reconstruye la conectividad estructural del cerebro, pero está limitada por la suposición de funciones de distribución de orientación de fibras (fODFs) simétricas.
  • Esta simetría forzada puede dificultar el modelado preciso en regiones cerebrales con microestructuras complejas y asimétricas de la sustancia blanca.

Objetivo del estudio:

  • Desarrollar y validar un marco de aprendizaje profundo no supervisado, Recursive-a-fODF, para la estimación directa de fODFs asimétricas (a-fODFs) a partir de datos de dMRI.
  • Eliminar la necesidad de conocimientos anatómicos incorporando un proceso de calibración recursivo basado en datos para estimar la función de respuesta de la sustancia blanca.
  • Demostrar la utilidad del modelado a-fODF para resolver configuraciones complejas de fibras e identificar alteraciones microestructurales específicas de la enfermedad.

Principales métodos:

  • Implementación de Recursive-a-fODF, un modelo de aprendizaje profundo no supervisado para estimar a-fODFs directamente a partir de dMRI.
  • Un proceso de calibración recursivo dentro del modelo estima dinámicamente la función de respuesta de la sustancia blanca a partir de los propios datos.
  • Validación utilizando conjuntos de datos de cerebro de tití ex vivo y cerebro humano in vivo, incluidas cohortes clínicas con afecciones neurodegenerativas y psiquiátricas.

Principales resultados:

  • Recursive-a-fODF demostró un rendimiento superior en la resolución de configuraciones complejas de fibras en comparación con los métodos convencionales.
  • La aplicación a cohortes clínicas reveló alteraciones específicas de la enfermedad en la asimetría de la orientación de las fibras, lo que pone de relieve la sensibilidad del método.
  • La estimación basada en datos de los a-fODF capturó con éxito firmas microestructurales relevantes para la patología de la enfermedad.

Conclusiones:
