Resumen

Este estudio muestra cómo los enlaces de halógeno y otros efectos del halógeno influyen en el autoensamblaje de péptidos. Los péptidos que contienen yodo presentan mayor estabilidad y mecanismos de autoensamblaje distintos en comparación con los análogos fluorados.

  • Los efectos del halógeno, más allá del XB directo, pueden impactar significativamente el comportamiento molecular.

Objetivo del estudio:

  • Investigar la modulación del autoensamblaje de péptidos mediante enlaces de halógeno (XB) y otros efectos del halógeno.
  • Comparar la influencia del yodo frente al flúor en péptidos funcionalizados sobre las propiedades de autoensamblaje.
  • Elucidar la interconexión entre XB, HB e interacciones hidrofóbicas en la autoorganización de péptidos.

Principales métodos:

  • Síntesis de tres pentapéptidos funcionalizados con un motivo FFK común.
  • Incorporación de una unidad de tetrafluoroiodofenilo (TFIP) como donante de XB.
  • Caracterización del autoensamblaje, la estabilidad térmica y las propiedades viscoelásticas mediante estudios exhaustivos.

Principales resultados:

  • El péptido funcionalizado con histidina mostró un enlace C-I···N significativo, mientras que el péptido terminado en fenilalanina mostró un enlace insignificante.
  • Los péptidos que contienen yodo exhibieron mayor estabilidad y mejores propiedades viscoelásticas que los análogos fluorados.
  • Se observaron mecanismos de autoensamblaje distintos (cooperativo vs. isodésmico), atribuidos a la hidrofobicidad y polarizabilidad del yodo.

Conclusiones:

  • Los enlaces de halógeno y otros efectos del halógeno, en sinergia con los enlaces de hidrógeno, pueden modular el autoensamblaje de péptidos.
  • Las propiedades únicas del yodo conducen a una mayor estabilidad y a comportamientos de autoensamblaje diferentes en comparación con el flúor.
  • Un delicado equilibrio de XB, HB e interacciones hidrofóbicas ajusta la autoorganización de péptidos.
