Resumen
La proteína 1 asociada a KRAB (KAP1) se sobreexpresa en el mesotelioma pleural (MP), promoviendo el crecimiento tumoral y la agresividad. KAP1 actúa como un regulador similar a un oncogén, afectando la proliferación celular, la migración y la invasión en el MP.

Sus antecedentes:
- El mesotelioma pleural (MP) es una neoplasia maligna agresiva con biomarcadores diagnósticos limitados.
- La comprensión de los mecanismos moleculares que impulsan la progresión del MP es crucial para el diagnóstico temprano y la terapia dirigida.
- El papel de la proteína 1 asociada a KRAB (KAP1) en la patogénesis del MP sigue sin explorarse en gran medida.
Objetivo del estudio:
- Investigar la expresión de KAP1 en tejidos de MP y su impacto funcional en el comportamiento de las células de mesotelioma.
- Evaluar KAP1 como un biomarcador diagnóstico potencial para la detección temprana de MP.

Principales resultados:
- KAP1 se sobreexpresó significativamente en tejidos de MP en comparación con tejidos pleurales normales.
- La sobreexpresión de KAP1 mejoró la proliferación, migración e invasión de las células MSTO-211H, aumentando los niveles de ciclina D1 y E.
- La reducción de KAP1 inhibió la proliferación, migración e invasión, indujo la detención del ciclo celular en fase G0/G1 y aumentó la apoptosis.
- La expresión de KAP1 se correlacionó positivamente con TP53 y SHOX2, y negativamente con MTAP y MSLN.
- El Análisis de Enriquecimiento de Conjuntos de Genes (GSEA) reveló un enriquecimiento asociado a KAP1 en las vías de reparación del ADN, ciclo celular y proteostasis.
Conclusiones:
- KAP1 se expresa en el mesotelioma pleural y actúa como un regulador similar a un oncogén.
