Activation of Integrins

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling."
In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.

La eferocitosis, el envolvimiento de las células apoptóticas, está mediada por complejos de receptores MERTK. Estos complejos involucran integrinas β2 y moléculas de señalización, revelando nuevos mecanismos para el aclaramiento celular y conocimientos sobre enfermedades inflamatorias.

Sus antecedentes:

  • La eferocitosis es crucial para la homeostasis tisular y la prevención de la autoinmunidad.
  • La tirosina quinasa del receptor MERTK media la eferocitosis en muchos tejidos.
  • Las vías de señalización precisas y los mecanismos moleculares de la eferocitosis mediada por MERTK siguen siendo en gran medida desconocidos.

Objetivo del estudio:

  • Aclarar los mecanismos moleculares y las vías de señalización que rigen la eferocitosis mediada por MERTK.
  • Para identificar los componentes proteicos de los complejos de receptores que contienen MERTK involucrados en la eferocitosis.

Principales métodos:

  • Se empleó la espectrometría de masas para identificar proteínas dentro de los complejos de receptores MERTK.
  • Se utilizó microscopía de súper resolución para visualizar la estructura y la dinámica de estos complejos durante la eferocitosis.
  • Los ensayos funcionales evaluaron las funciones de las integrinas MERTK y β2 en la envoltura celular apoptótica.

Principales resultados:

  • Se identificaron complejos de receptores de 180 nm que contienen MERTK, integrinas β2 y moléculas de señalización asociadas.
  • Se demostró que MERTK induce un cambio conformacional dependiente de la PI3-cinasa en las integrinas β2, promoviendo la unión de alta afinidad.
  • Se caracterizó por una sinapsis eferocítica altamente estructurada con MERTK en el centro y un anillo en expansión de integrinas β2 y actina en el borde delantero, dependiente de las quinasas de la familia Src y FAK.

Conclusiones:

  • Las integrinas MERTK y β2 cooperan para mediar la eferocitosis a través de una vía de señalización estructurada.
  • Los hallazgos proporcionan nuevos conocimientos sobre la función de MERTK en el aclaramiento celular.
  • Comprender estos mecanismos puede iluminar la contribución de los defectos de MERTK a las enfermedades inflamatorias y autoinmunes.
