    Sus antecedentes:

    • La clonación T/A es un método ampliamente utilizado para crear plásmidos de ADN recombinante.
    • Esta técnica utiliza pares de bases simples de adenina-timina (A:T) para la ligación de insertos en vectores.
    • Teóricamente, la orientación del inserto durante la ligación es aleatoria.

    Objetivo del estudio:

    • Investigar el sesgo de direccionalidad observado en la clonación T/A utilizando el Sistema de Vectores pGEM-T.
    • Cuantificar el alcance de este sesgo en un entorno de Experiencia de Investigación de Pregrado Basada en Cursos (CURE).
    • Destacar las implicaciones prácticas de este sesgo para la eficiencia de la clonación molecular.

    Principales métodos:

    • Se utilizó el Sistema de Vectores pGEM-T para la clonación T/A.
    • Se realizaron clonaciones de más de 400 insertos de ADN.
    • Se analizaron los productos de ligación para determinar la orientación del inserto en relación con el promotor T7.

    Principales resultados:

    • Se observó consistentemente un sesgo de direccionalidad significativo en la ligación de insertos.
    • El sesgo favoreció la orientación "sentido" hacia el promotor T7 en más del 74% de los clones.
    • El análisis estadístico confirmó la significancia de este sesgo (valor p < 0.0005).

    Conclusiones:

    • La clonación T/A con el Sistema de Vectores pGEM-T exhibe un sesgo de direccionalidad predecible.
    • Comprender este sesgo es crucial para optimizar las estrategias de clonación.
    • La conciencia de este fenómeno puede reducir el tiempo y los costos experimentales en la investigación en biología molecular.
