  • Demostrar la utilidad de la plataforma para la construcción de vectores de baculovirus avanzados para la edición génica.

Principales métodos:

  • Desarrollo de SHOT 2.0, un sistema de transposón CRISPR-asociado para la edición de bacmids guiada por ARN en Escherichia coli.
  • Integración sitio-específica de cargas de ADN grandes (≥14 kb) en loci de bacmid (v-cath, ODVe56).
  • Construcción y prueba de un baculovirus todo en uno que codifica un editor primario (PE5max).
  • Evaluación de la eficiencia de edición primaria en células HEK293T, células madre pluripotentes inducidas (iPSC) y células de cáncer de hígado.

Principales resultados:

  • SHOT 2.0 permite la integración sitio-específica de cargas de ADN grandes (≥14 kb) en loci de bacmid definidos.
  • La integración en el locus ODVe56 mejora significativamente la estabilidad del transgén durante el pasaje serial de virus.
  • El sistema es compatible con el flujo de trabajo Bac-to-Bac®, permitiendo la inserción de genes duales.
  • La edición primaria mediada por vector alcanzó una eficiencia de hasta 85.6% en células HEK293T y 37.1% en iPSC y células de cáncer de hígado.

Conclusiones:

  • SHOT 2.0 expande significativamente la caja de herramientas de ingeniería de baculovirus, ofreciendo una plataforma flexible para la edición del genoma.
  • La plataforma facilita la creación de vectores de baculovirus avanzados para diversas aplicaciones, incluida la entrega de genes.
  • Se demostraron capacidades robustas de edición primaria en varios tipos de células, incluidas aquellas desafiantes como iPSC y células cancerosas.
