Sus antecedentes:

  • Los condensados anclados por ARN son cruciales para las funciones celulares y las enfermedades.
  • El mapeo de la arquitectura de proteínas dentro de estos condensados a resolución subcompartimental es un desafío.

Objetivo del estudio:

  • Desarrollar una tecnología novedosa para el mapeo de alta resolución de proteomas proximales a ARN.
  • Investigar la organización espacial y la partición de proteínas dentro de los subcompartimentos nucleolares.

Principales métodos:

  • Se introdujo HCR-Proxy, una técnica de marcado de proximidad que combina la amplificación de la señal de reacción en cadena de hibridación (HCR) con la biotinailación in situ de proximidad (Proxy).
  • Se aplicó HCR-Proxy a objetivos de pre-ARN nacientes en el nucléolo.
  • Se integraron los resultados con marcos de aprendizaje profundo.

Principales resultados:

  • HCR-Proxy permitió el perfilado proteómico de proteomas proximales a ARN a resolución subcompartimental.
  • Reveló firmas proteómicas distintas de los subcompartimentos nucleolares.
  • Descubrió la lógica espacial impulsada por la secuencia de ARN de la partición de proteínas basada en la carga, el peso molecular y los dominios de unión a ARN.

Conclusiones:

  • HCR-Proxy proporciona mapas resueltos espacialmente de interactomas de ARN dentro del nucléolo.
  • Demostró una base codificada por secuencia para la organización de proteínas en subcompartimentos de condensados anidados.
  • Estableció HCR-Proxy como una plataforma escalable para el descubrimiento de interactomas de ARN, vinculando la localización de transcritos con el contexto proteómico.
