Sus antecedentes:

  • La homeostasis ósea depende de la intrincada comunicación entre las células óseas y el tejido adiposo dentro de la médula ósea.
  • Un desequilibrio que favorece el desarrollo de las células grasas (adipogénesis) sobre el desarrollo de las células óseas (osteogénesis) se asocia con la osteoporosis y el aumento de la grasa medular.
  • Se postula que los factores derivados de los adipocitos modulan la actividad de los osteoblastos y el metabolismo óseo.

Objetivo del estudio:

  • Investigar el impacto del medio condicionado por adipocitos (CM) en la función y diferenciación de los osteoblastos.
  • Identificar los cambios moleculares en los osteoblastos expuestos a secreciones de adipocitos.
  • Elucidar los mecanismos por los cuales los adipocitos influyen en el equilibrio osteo-adipogénico.

Principales métodos:

  • Tratamiento de osteoblastos con medio condicionado derivado de adipocitos.
  • Evaluación de la viabilidad celular, la actividad de la fosfatasa alcalina (ALP) y la deposición de calcio.
  • Análisis proteómico para identificar proteínas expresadas diferencialmente.
  • Análisis bioinformático que incluye análisis de redes STRING y evaluación de marcadores epigenéticos.

Principales resultados:

  • El CM de adipocitos no causó citotoxicidad pero alteró significativamente la diferenciación osteoblástica, evidenciado por la reducción de la deposición de calcio y la actividad de la ALP.
  • El análisis proteómico reveló la regulación descendente de proteínas cruciales de la matriz extracelular y la regulación ascendente de proteínas asociadas con la inflamación y el estrés oxidativo (por ejemplo, GAPDH, vimentina).
  • El análisis de redes indicó un enriquecimiento en las vías de respuesta al estrés, mientras que las alteraciones epigenéticas como la sobreexpresión de hemoglobina y la reducción de isoformas de histonas sugirieron la activación de NF-κB y el estrés oxidativo.

Conclusiones:

  • El secretoma de adipocitos crea un microambiente proinflamatorio y rico en estrés oxidativo que es perjudicial para la diferenciación y función de los osteoblastos.
  • La mineralización alterada, la disfunción del citoesqueleto y las modificaciones epigenéticas adversas son consecuencias clave de la comunicación entre adipocitos y osteoblastos.
  • La focalización de estas vías moleculares puede ofrecer nuevas estrategias terapéuticas para la osteoporosis y trastornos óseos relacionados.
