La citometría de flujo puede detectar varias células inmunitarias en la saliva, revelando distintas respuestas inmunitarias mucosas. Este método ofrece una nueva forma de estudiar enfermedades que afectan la inmunidad mucosa.

Sus antecedentes:
- La cavidad oral es una parte clave del sistema inmunitario mucoso del cuerpo, interactuando con numerosos antígenos.
- Si bien los mediadores solubles de la inmunidad mucosa están bien estudiados, los fenotipos de las células inmunitarias siguen siendo menos comprendidos, lo que dificulta la investigación de la patogénesis de las enfermedades.
Objetivo del estudio:
- Revisar la importancia del fenotipado inmunitario en las secreciones mucosas.
- Demostrar la utilidad de la citometría de flujo para cuantificar diversos fenotipos de linfocitos en el fluido bucal estimulado (SWMF).
Principales métodos:
- Se realizó una revisión de las técnicas de fenotipado inmunitario.
- Se utilizó la citometría de flujo para detectar y cuantificar fenotipos de linfocitos (Th17, Th22, Tfh, Tregs, NKT, NK, ILC1, ILC2, ILC3) en SWMF y células mononucleares de sangre periférica (PBMC) de 119 participantes.
Principales resultados:
- La citometría de flujo detectó de manera confiable varios fenotipos de células inmunitarias en SWMF.
- Las frecuencias de células Th17, Th22, Tfh y NK fueron mayores en SWMF que en PBMC.
- Las correlaciones de fenotipos sugirieron un origen mucoso específico para las células detectadas en SWMF.
Conclusiones:
- La citometría de flujo es eficaz para detectar una amplia gama de fenotipos de linfocitos en SWMF.
- Esta técnica es prometedora para investigar la inmunopatogénesis de enfermedades que afectan los sitios mucosos.
