Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

    Principales métodos:

    • Diseño y generación de bioPROTACs para aplicaciones en células CAR T.
    • Expresión de bioPROTACs en células CAR T dirigidas a DNMT3A.
    • Evaluación de los efectos funcionales de la expresión de bioPROTAC en el fenotipo de las células T.

    Principales resultados:

    • La expresión de bioPROTAC en células CAR T dirigidas a DNMT3A fenocopió los efectos de la eliminación génica.
    • El enfoque bioPROTAC demostró una estrategia reversible y sintonizable.
    • Este método reprogramó con éxito el destino de las células T, abordando el agotamiento de las células T.

    Conclusiones:

    • Los bioPROTACs diseñados de novo proporcionan una alternativa viable que no implica edición génica para mejorar la función de las células CAR T.
    • Este enfoque reversible ofrece un control sintonizable sobre la reprogramación del destino de las células T.
    • Los bioPROTACs tienen una amplia aplicabilidad para el desarrollo de terapias celulares de próxima generación.
